Structure-based Design, Synthesis and Evaluation of Inhibitors of Trypanothione Reductase

Trypanothione Reductase

Human African Trypanosomiasis (HAT), better known as sleeping sickness, is a parasitic tropical disease found in Southeast and West Africa that causes around 50,000 deaths per year. HAT is transmitted by the Tsetse fly and it is caused by the parasite T. Brucei. The enzyme trypanothione reductase (TR) is a crucial component for the protection of the parasite from oxidative stress and inhibition of the enzyme leads to the parasite's death. Therefore, TR represents an highly attractive target for the treatment of human African Trypanosomiasis. The aim of this project is the design, synthesis and biological evaluation of novel TR inhibitors based on the knowledge obtained in former investigations.
